เจ้าฟ้าหญิงมาเรียแห่งโรมาเนีย ประสูติเมื่อวันที่ 13 กรกฎาคม พ.ศ. 2507 ที่เมืองเฮลเลรับ ประเทศเดนมาร์ก เป็นพระธิดาองค์สุดท้องใน สมเด็จพระราชาธิบดีไมเคิลที่ 1 แห่งโรมาเนียกับสมเด็จพระราชินีแอนน์แห่งโรมาเนีย
ในวันที่ 16 กันยายน พ.ศ. 2538 ในเมืองนิวยอร์ก ประเทศสหรัฐอเมริกา เจ้าฟ้าหญิงมาเรียทรงอภิเษกสมรสกับคาซิมีร์ มิสคอว์สกี ชาวโปแลนด์ซึ่งเป็นนิกายโรมันคาทอลิก ทำให้เจ้าฟ้าหญิงมาเรียทรงสูญเสียลำดับการสืบราชบัลลังก์อังกฤษ ทั้งคู่ทรงหย่าร้างในปีพ.ศ. 2546 ทรงไม่มีพระโอรสพระธิดา
